Calcium Leucovorin Injection, also known as folinic acid calcium, is a form of reduced folic acid used medically as a “rescue” agent during high-dose methotrexate chemotherapy. Methotrexate inhibits dihydrofolate reductase, blocking folic acid metabolism and DNA synthesis, which affects both cancerous and normal cells. Leucovorin bypasses this blockade by providing an active form of folate, helping to rescue normal cells from methotrexate toxicity.

In addition to methotrexate rescue, Calcium Leucovorin is used to treat folic acid deficiency due to malnutrition, certain anemias, or vitamin deficiencies. It also enhances the efficacy of 5-fluorouracil (5-FU) chemotherapy by stabilizing its binding to the target enzyme thymidylate synthase.

This injection is administered intravenously or intramuscularly under medical supervision, with dosage depending on the clinical indication and patient response. It is essential for preventing or mitigating side effects such as mucositis, bone marrow suppression, and neurotoxicity associated with methotrexate therapy.
